Is it possible to have a simple % increase on standard rates?
eg.
Normal working hours = 0%
20.00 to 00.00 hours = +10%
00.00 to 06.00 hours = + 20%

Or is it necessary to set up all the hours for each day separately?

you can use Tariffs to increase the price for different time intervals during the day: https://booking.drivenot.com/en/documentation/46-tariffs
This method only works for Address to Address (taxi style) per mile set up.

If you are using Fixed fare routes then you will have to create different Cars with different Availability times and then assign different price for each car in Fixed fare route Edit screen.
